چکیده

The reversible transformation of galactose into glucose in biological systems is important both for the utilization of dietary galactose as fuel (via hexose 6-phosphate) and for the biosynthesis of complex galactose-containing compounds from glucose or acetate. The first step in the metabolism of galactose in both yeast and animal tissues is phosphorylation in the 1 position of the hexose to form cw-Gal-l-P’ (l-3). The interconversion of the hexoses then takes place according to the following equations, in which uridine diphosphoglucose functions catalytically.

Practical Enzyme-Based Syntheses of Uridine 5'-Diphosphogalactose and Uridine 5'-Diphospho-/V-acetylgalactosamine on a Gram Scaler

Practical enzyme-based routes for the syntheses of uridine 5'-diphosphogalactose (UDP-Gal) and uridine 5'-diphospho-N-acetylgalactosamine (UDP-GalNAc) on millimole scales have been developed. The activity of galactokinase (EC 2.7.L.6) in crude enzyme extracts from galactose-adapted yeast, coupled to a regenerating system for ATP, provides convenient and economical access to galactose-a-1-phosph...

We wish to thank Karen Rutherford, Erna Rollefson, Charles Hill, and Douglas MacPherson for assistance in various phases of these experiments, and Julius Adler for the Lederberg mutants and helpful comments. The following abbreviations are used: Gal, galactose; Gal-1-P, a-D-galactose-1-phosphate; G-1-P, a-D-glucose-l-phosphate; G-6-P, glucose-6-phosphate; UDPGal, uridine diphosphogalactose; UDP...

Issues on universal screening for galactosemia.

Galactosemia is an inborn error of galactose metabolism, caused by an abnormality in the conversion of galactose and uridine diphosphoglucose to glucose-1-phosphate and uridine diphosphogalactose through the action of 3 sequential enzymes: galactokinase (GALK), galactose- 1-phosphate uridyltransferase (GALT), and uridine phosphogalactose 4-epimerase (GALE). The advent of newborn screening broug...

Uridine diphosphoacetylglucosamine pyrophosphorylase.

This mechanism, termed pyrophosphorolysis, was first observed by Kornberg (1, 2) who investigated the pyrophosphorolytic cleavage of diphosphopyridine nucleotide, and the reverse reaction between adenosine triphosphate and nicotinamide ribonucleotide resulting in diphosphopyridine nucleotide synthesis. Since that time, enzymes which catalyze the synthesis of flavin adenine dmucleotide (3), deph...

Phosphoglucomutase mutants of Escherichia coli K-12.

Bacteria with strongly depressed phosphoglucomutase (EC 2.7.5.1) activity are found among the mutants of Escherichia coli which, when grown on maltose, accumulate sufficient amylose to be detectable by iodine staining. These pgm mutants grow poorly on galactose but also accumulate amylose on this carbon source. Growth on lactose does not produce high amylose but, instead, results in the inducti...
